Fiona Donnison, 43, was arrested on suspicion of murder following the discovery in Heathfield, East Sussex, on Wednesday morning.
She was held after visiting Heathfield police station 'suffering from some distress' to report 'an incident', Sussex Police said.
An ambulance and two response cars were sent and officers found the bodies of Harry and Elise, both aged under five, after forcing entry into a silver Nissan.
Both children were confirmed dead at the scene.
Ms Donnison, who is awaiting questioning, was taken to Eastbourne District General Hospital with 'apparent self-harm injuries' to her arms and is now in a stable condition.
The car was towed away for forensic examination as a police cordon was put in place around a nearby three-storey detached house.
Former neighbour Jill Radford, 61, said: 'I think there used to be a couple living there until they separated. The house went up for sale some time before Christmas.'
Chief Superintendent Robin Smith said police could not say how the children had died but officers were not looking for anyone else in connection with the deaths.
The inquiry is being led by Sussex Police but the force has been in contact with officers in Surrey, where Ms Donnison is believed to be from.
'We believe she lives in Surrey but has connections to Heathfield. We believe the children have been residents in Heathfield in the past,' said Ch Supt Smith.
Family members have been informed of the deaths, he said.
Ch Supt Smith added: 'It's a huge tragedy for all involved, and not only for the family - it has a huge impact on the local community.
Ms Donnison, a former Lloyds syndicate worker in the City, also has two teenage sons.
